1.Moisturizing Efficacy

Test Objective: This test establishes an *in vitro* protocol to evaluate the moisturizing efficacy of cosmetic products and raw materials by measuring their water-retention performance in a dry environment.

*In Vitro* Moisturizing Efficacy Evaluation: Three replicates were set up for each test group. Negative and positive control groups were included alongside the test groups; glycerin served as the positive control, and purified water as the negative control. The procedure for the controls was identical to that for the test samples: A clean, dry, stoppered glass weighing bottle was placed in a desiccator (containing indicating silica gel desiccant) at 25°C ± 1°C for 24 hours prior to the test, and its weight (m0) was accurately recorded. At room temperature, 1 mL of nutrient agar was pipetted into the weighing bottle; after the agar solidified, 200 μL of the test sample was added, and the total mass (m1) was accurately recorded. The bottles were then placed in a desiccator containing desiccant for a total duration of t = 8 hours. For both the test and control groups, the weighing bottles were removed from the desiccator at 2, 4, 6, and 8 hours; the stoppers were secured, and the weight (mt) was accurately recorded. Moisturizing efficacy was evaluated based on the percentage of weight loss due to drying.

Test Conclusion: Under the conditions of this test, the results showed that the percentage weight loss for the sample group at 2, 4, 6, and 8 hours was lower than that of the negative control group, indicating that the sample possesses moisturizing properties.

2.Repair

Test Protocol: This test evaluated the repair efficacy claims of the submitted sample by measuring specific parameters in 30 healthy Asian (Chinese) subjects (male and female) before and after product application. The study included 30 valid subjects, achieving a 100% validity rate.

 

Skin transepidermal water loss (TEWL) value
2 hours after using the sample The rate of change in transepidermal water loss (TEWL) for the skin in the test area among the 30 subjects was -30.63%, while that for the control area was -23.03%; a significant difference was observed between the two groups.
4 hours after using the sample Among the 30 subjects, the rate of change in transepidermal water loss (TEWL) for the skin in the test area was -35.00%, while that for the control area was -24.34%; a significant difference was observed between the two groups.
conclusion The results indicate that the test samples exhibit repair efficacy after 2 and 4 hours of use.
Skin Hemoglobin EI Value
2 hours after using the sample The rate of change in the skin hemoglobin EI value in the test area for the 30 subjects was -16.54%, while that in the control area was -12.26%; there was no significant difference between the two groups.
4 hours after using the sample The rate of change in the skin hemoglobin EI value in the test area for the 30 subjects was -22.18%, while that in the control area was -16.09%; there was no significant difference between the two groups.

 

Statistical analysis was performed using SPSS 26.0 to evaluate the validity of the test parameters. The test data were subjected to a normality test; data following a normal distribution were analyzed using the t-test, while non-normally distributed data were analyzed using the rank-sum test. A significance level of p < 0.05 was adopted, with statistical significance defined within a 95% confidence interval.

Regarding the correlation between the test parameters and the efficacy claim, “repair” refers to the maintenance of the treated area in a normal state; therefore, the repair efficacy of the sample can be assessed by measuring improvements in Transepidermal Water Loss (TEWL), stratum corneum water content, or the skin hemoglobin Erythema Index (EI) before and after product application.

Measurement of stratum corneum water content is based on the internationally recognized capacitance method, which assesses changes in skin capacitance before and after application to determine if stratum corneum water content has increased; a higher measured value indicates higher stratum corneum water content following sample application. The TEWL value serves as an indicator of skin barrier function; a lower TEWL value signifies reduced transepidermal water loss and superior skin barrier function. A lower measured skin hemoglobin EI value indicates a lower hemoglobin content. The sample is considered to possess repair efficacy if, following application, there is a significant decrease in TEWL and a significant increase in stratum corneum water content, or a significant decrease in the skin hemoglobin EI value.
