Study Protocol: A total of 30 Asian (Chinese) male and female subjects aged 22–60 were recruited for the test (30 subjects enrolled, 30 completed the study; completion rate: 100.00%). Test Methods:

① Anti-wrinkle and firming – Elastase inhibition rate: Samples are diluted with TESCA buffer solution according to specified ratios. Components are then added as shown in the table below, dividing the setup into: Well 1 Test Group (sample group and positive control group), Well 2 Test Control Blank Group, Well 3 Negative Control Group, and Well 4 Negative Control Blank Group. Upon completion of the reaction, the reaction mixtures are transferred to the corresponding 96-well plates (with three replicates per group), and absorbance (OD value) is measured at 410 nm using a microplate reader.

② Hyaluronidase inhibition rate: Components are added to the sample group and positive control group, both of which have been diluted with purified water according to specified ratios.

③ In vitro evaluation of moisturizing efficacy: Three replicates are set up for each test group. Negative and positive control groups are included simultaneously; glycerin is used for the positive control group, and purified water for the negative control group. The procedure follows that of the sample group: a dry, stoppered glass weighing bottle is placed in a desiccator (containing indicating silica gel desiccant at the bottom) at 25°C ± 1°C for one day prior to the test, and its weight (m0) is accurately recorded. At room temperature, 1 mL of nutrient agar is pipetted into the weighing bottle; after solidification, 200 μL of the test sample is added, and the total mass (m1) is accurately recorded. The bottle is then placed in the desiccator containing desiccant for a total duration of t = 8 hours. For both the sample and control groups, the weighing bottle is removed from the desiccator at 2, 4, 6, and 8-hour intervals; the lid is secured, and the weight (mt) is accurately recorded.

Moisturizing performance was evaluated based on the percentage of weight loss due to drying. Test conclusions:

① Under the test conditions, the elastase inhibition rate of the sample (BONNEHEUR Recombinant Collagen & Peony Repairing Anti-Wrinkle Face & Neck Cream, at a concentration of 20 mg/mL) was 61.4% ± 1.9% (P < 0.05, indicating a significant difference). ② Under the test conditions, the hyaluronidase inhibition rate of the sample (BONNEHEUR Recombinant Collagen & Peony Repairing Anti-Wrinkle Face & Neck Cream, at a concentration of 20 mg/mL) was 69.8% ± 1.4% (P < 0.05, indicating a significant difference). It is concluded that the sample inhibits hyaluronidase activity, demonstrating soothing properties.

③ Under the test conditions, the test results showed that the weight loss percentages for the sample group at 2, 4, 6, and 8 hours were all lower than those of the negative control group, indicating that the sample (BONNEHEUR Recombinant Collagen & Peony Repairing Anti-Wrinkle Face & Neck Cream) possesses moisturizing properties.
