Therefore, the repair efficacy of a sample can be evaluated by measuring improvements in these parameters—specifically TEWL rates, hemoglobin (EI) values, and stratum corneum moisture content—in subjects before and after using the sample.
A lower TEWL value indicates reduced transepidermal water loss; similarly, a lower hemoglobin (EI) value combined with higher stratum corneum moisture content indicates superior skin barrier function, signifying that the sample possesses repair efficacy.
Moisturizing
This test establishes an *in vitro* protocol to assess water-retention capabilities, thereby evaluating the moisturizing efficacy of cosmetic products and raw materials by measuring their ability to retain moisture in a dry environment.
Test Method: Three replicates were set up for each test group. Negative and positive control groups were included alongside the test groups; the positive control used glycerol, while the negative control used purified water. The procedure was identical for all groups: A clean, dry, glass weighing bottle with a stopper was placed in a desiccator (containing indicating silica gel desiccant) at 25°C ± 1°C for 24 hours prior to the test, and its weight (m0) was accurately recorded. At room temperature, 1 mL of nutrient agar was dispensed into the weighing bottle; after solidification, 200 μL of the test sample was added, and the total mass (m1) was accurately recorded. The bottles were then placed in a desiccator containing desiccant for a total duration of t = 8 hours. For both the sample and control groups, the weighing bottles were removed from the desiccator at 2, 4, 6, and 8-hour intervals; the lids were secured, and the weight (mt) was accurately recorded.
